What is the thread size of the barb metal connector - 1/4 in. or 3/8 in?

The hose Barbs appears to be molded into the housing, for the supplied 1/8" ID dia hose.

why are there 2 connector ports?

Second port is for measuring differential pressure, not needed if only reading pressure, and there is a positive and negative.

Why does it read -.018"H2O without even taking the measurement? That would be quite an error.

Because it hasn’t been “zero’d” yet.
