does canister self seal, can you you remove burner then put back on, Sorry for obvious?

Yes. I have use a single times, each time disconnecting it.

can i refill this, if so where could i get it done and for how much?

As far as I know these canisters are not refillable.

Can I attach and detach this from my burner? Or do they have to stay connected once I screw them together?

You can detach the fuel canister after you are finished cooking and store it inside the pot with all the other things, burner, cup for easy packing and storage.
