Q: Will this bag fit under the seat or in the overhead compartment on a regional/commuter jet? I am worried about having to gate check it.

If you can get it under the seat in front of you, I'd be surprised. It depends on the aircraft. I've flown in many Embraer RJ145s. You should be able to get a messenger bag in the stowage compartment above, but not this bag.

Q: can wheels be removed? In case it doesn't fit in an airplane overhead.

NO - but I have been flying domestic with this for years and it always fits - if you fly international it is too big for overhead anyway :)

Q: Has anyone ever had to check this bag when it didn't fit on a smaller airplane? If so, did it still keep your gear safe?

I have had multiple occasions what was necessary To check the bag at the gate when boarding a small commercial plane. I have lost nothing. The bag will lock with the key and lock.
