Hi, Good question. I'll have to go measure one to get an exact dimension for you. The piece that comes apart is the grey piece with tabs that grips the line. The line sits pretty close to the edge of the spool, within the grey gripper. The gripper has two sides with slightly different grip. Q: Can this be used for a mono rig, a fly leader with a base of 20 lb Maxima Chameleon and an overall length of 25-30 feet?

Should work fine I've used it with a mono tenkara line

Q: Looks like this spool will not slide over the end of a tenkara rod like more basic tenkara line spools will. am i correct in assuming this?

The larger hole slides on the grip of my Tenkara USA Rhodo rod. I put it on with the rod collapsed

Q: Are the flies and line included?

Nothing is included. You get basically a carrying case for the line and fly for you rod. I use it for my 720 Tenkara rod. For my smaller backpack rods I use line clamps that hold the line and fly. Tenkara line clamps are available on Amazon.
