Yes, they can be worn with eye glasses. The Ortopad eye patches are designed with nasal slits to allow the patch to lie flat against the side of the nose.

Is it safe to keep on over night?

The adhesive is about as strong as a band aid, albeit a good one. We normally take them off of our 6 year old at bedtime.

No. They are disposable. They seem to fit better than anything else I've found (disposable or not). The non-disposable ones that fit over the glasses don't seem to be 100% light blocking so our specialist said they are basically ineffective if you are treating lazy eye. Which was disappointing because my daughter gets very sweaty when she's active and then these fall right off. I get about 2 hours wear out of each one then have to replace. But these were the ones they recommended and give out in the office as well.
